Because of the drawbacks related with the current corneal transplantation techniques such as donor shortage, rejection or disease transmission risk, there is still an urgent requirement in order to produce new treatments for regenerating corneal tissue. Specially, it is also necessary to overcome the problems related with the blindness and vision losses originating from damage to corneal endothelium. By considering these, corneal tissue engineering might possess an encouraging potential to ensure alternative and innovative approaches toward the development of effective corneal tissue equivalents. Peptides allow to the creation of bi . . .omimetic and bioactive material surfaces, thus peptide-modified scaffolds have become a popular subject of research in tissue engineering. Herein, our objective was to synthesize the corneal endothelium tissue-specific peptides, to conjugate these peptides on created polyvinyl alcohol (PVA)-xanthan gum (XG) membranes, to characterize the peptide conjugation and also to define the influence of those peptide conjugated structures on cellular viability to utilize in corneal endothelium tissue engineering. Mevcut kornea transplantasyonu teknikleriyle ilgili donör eksikliği, ret veya hastalık bulaşma riski gibi dezavantajlar nedeniyle, kornea dokusunun rejenerasyonu için yeni tedavilerin üretilmesi için hala acil bir ihtiyaç bulunmaktadır. Özellikle, kornea endoteli hasarından kaynaklanan körlük ve görme kayıplarına bağlı problemlerin de üstesinden gelinmesi gerekmektedir. Bunları göz önünde bulundurarak, kornea dokusu mühendisliği, etkili kornea dokusu eşdeğerlerinin geliştirilmesine yönelik alternatif ve yenilikçi yaklaşımlar sağlama konusunda umut verici bir potansiyele sahip olabilir. Peptitler, biyomimetik ve biyoaktif malzeme yüzeylerinin oluşturulmasına izin verir, bu nedenle peptid ile modifiye edilmiş yapı iskeleleri, doku mühendisliğinde popüler bir araştırma konusu haline gelmiştir. Burada, amacımız, kornea endoteli dokusuna özgü peptitlerin sentezlenmesi, bu peptitlerin oluşturulan polivinil alkol (PVA)- ksantan sakızı (KS) membranları üzerine konjuge edilmesi, peptit konjugasyonunun karakterize edilmesi ve ayrıca kornea endotel doku mühendisliğinde kullanım için bu peptit konjuge yapıların hücresel canlılık üzerindeki etkisinin tanımlanmasıdır. İlk olarak, geliştirilen PVA-KS membranlar üzerindeki peptit konjugasyonunu doğrulamak için, peptit konjuge numuneler Taramalı Elektron Mikroskobu (TEM), Enerji Dağılımlı X-Işını Spektroskopisi (EDX), Fourier Dönüşümlü Kızılötesi Spektroskopisi (FTIR) ve X-ışını Fotoelektron Spektroskopisi (XPS) analizleri ile karakterize edildi. Mevcut çalışmanın karakterizasyon sonuçları, sentezlenen peptitlerin PVA-XG membranlara başarıyla konjuge edildiğini gösterdi. Ayrıca, bu peptit ile modifiye edilmiş membranların insan kemik iliğinden türetilmiş mezenkimal kök hücrelerin (iMKH'ler) canlılığı ve çoğalması üzerindeki etkisi, MTT analizi ve Canlı/Ölü boyama tahliline göre değerlendirildi. Hücre canlılığı deneylerinin bulguları, geliştirilen peptit konjuge membran yapılarının iMKH'ler üzerinde toksik bir etkiye neden olmadığını ortaya koydu. Genel olarak, bu çalışmanın sonuçları, biyoaktif peptit modifiye PVA-XG membranların, kornea endoteli doku mühendisliği için kullanılma potansiyeline sahip olduğunu göstermiştir. The effect of RGD pertide on cell proliferation and vasculogenesis during microtissue formation

3D (Dimensional) microtissues are commonly used in regenerative medicine, medicine design, toxicology studies and advance cancer research. Microtissues which are constructed by this technique mimic natural in vivo condition and it stands for shifting form between in vitro studies and in vivo research. Scaffold-free microtissues produce own extracellular matrix and cells proliferate faster. However microtissue is used in vitro experiments, they mimic in vivo condition by producing own matrix. This peptide has been shown to enhance cell proliferation by increasing cellular interaction and cell-matrix connection in previous studies. HUVEC (Human Umbilical Vein Endothelial Cell) is one of the most commonly used model cells to understand the endothelial cell behavior. Moreover, the inner layer of the vascular structure is composed of endothelial cells. HUVEC is one of the model cell types frequently used in vascularization studies with high vasculogenesis capacity and endothelial origin. VEGF, Tie-1, Tie-2, PECAM, and Ve-cadherin are also effective genes for expressing preliminary vascular model studies in microtissue studies. This study was carried out to investigate the effect of RGD peptide on HUVEC scaffold-free 3D microtissue formation and early vasculogenesis. Microtissue size analysis, viability analysis, focal adhesion analysis and qRT-PCR (Real Time PCR) data were used. In the results of working, we found that RGD peptides increase HUVEC microtissue proliferation and contribute to constituting microtissue formation. Furthermore, it plays the effective role of HUVEC 3D microtissue formation during vascularization process. We experimented different RGD concentration on microtissue formation. Today, tissue transplant applications are widely used for repair of damaged hard tissue. Despite the gold standard of autografts, interest in synthetic bone grafts produced by tissue engineering techniques is increasing day by day, due to limiting factors such as damage to the tissue site and limited graft availability in allografts, and the risk of developing an immune system response in allografts.Many bioceramic materials, including β-tricalcium phosphate (β-TCP), hydroxyapatite (HA) and calcium sulfate, are widely used in bone tissue engineering. β-TCP has been the most preferred bioceramics in recent years due to its high osteo . . .-compatibility, fast degradation rate and high mechanical strength. A major disadvantage of existing implant materials is their sintered solid and hard form, which makes it difficult for the surgeon to adapt the surgical graft material to the desired shape during surgery. This causes bone loss and trauma to healthy peripheral tissues and prolonged surgical time. Polymer-ceramic based composite scaffolds are produced to overcome this problem. It is aimed to increase cell adhesion by mimicking the extra cellular matrix with its polymeric character and to imitate bone structure with ceramic character and show osteoconductive and osteoinductive properties. Within the scope of this study, chitosan based scaffolds with different β-TCP ratios were prepared by freeze drying method. For characterization of the scaffolds produced, morphological characteristics were investigated by Scanning Electron Microscope (SEM) and pore diameters were calculated. At the same time, compression test was performed to determine the mechanical properties. The biocompatibility of the produced scaffolds was supported by in vitro cytotoxicity and genotoxicity tests. Bone marrow-derived mesenchymal stem cells were used and cultured on scaffolds for the investigation of the effect of β-TCP content on stem cell differentiation. SERAMİK TABANLI 3B DOKU İSKELELERİNİN GELİŞTİRİLİP, ÜRETİLMESİ VE KARAKTERİZASYONU ÖZET Defekte uygun doku iskelesi üretimi kişiye özel yapay kemik uygulamalarında en kritik basamağı oluşturmaktadır. Üç boyutlu (3B) yazdırma özellikle kişiye özel uygulamalarda iç ve dış tasarımın yüksek kontrol edilebilirliği ile en iyi üretim tekniklerinden biridir. Bu çalışma kapsamında kemik doku mühendisliğine iyi bir alternatif oluşturmak üzere çinko katkılı, seramik temelli (kalsiyum sülfat (KS)) doku iskelelerinin 3B yazdırma ile geliştirilip üretilmesi ve karakterizasyon testlerinin yapılması amaçlanmıştır. 3B tasarlanan iskeleler 3B y . . .azıcı ile üretilmiştir. Çinko katkılı ve katkısız KS içerikli iskele grupları XRD, temas açısı ve SEM analizleri ile karakterize edilmiştir. Mekanik özellikleri basma testi yapılarak değerlendirilmiştir. Hücre canlılığı, MTT testi ile hücrelerin yüzeye tutunma yeteneği ise SEM gözlemleri ile belirlenmiştir. XRD analizi 3B yazıcının ticari tozu olan kalsiyum sülfat hemihidratın (KSHH), yapıştırıcı solüsyonun (binder) yüksek su içeriği nedeniyle yazdırma işlemi sonrası gypsuma dönüşmüş olduğunu, sinterleme işlemi sonunda ise su kaybı nedeni ile anhidrit formuna dönüşmüş olduğunu göstermektedir. Temas açı ölçümleri KSHH’ın hidrofilik karakterde olduğu ve ayrıca çinko ilavesinin KSHH örneklerin mekanik özelliklerini ve BMSC hücre hattının KSHH örnekler üzerindeki canlılığını olumlu yönde etkilediği görülmüştür. Benzer şekilde, SEM görüntülerinde görüldüğü üzere hücreler yüzeyde yassılaşarak oldukça iyi bir tutunma göstermektedirler. Sonuç olarak %0.5 çinko katkılı örneklerin kontrol, %0.1, %0.3 ve %0.5 çinko katkılı gruplar arasında en yüksek mekanik dayanıma sahip olduğu ve hücre canlılığı üzerindeki pozitif etkisinin de en fazla olduğu görülmüştür. xxiv Çalışmanın uzun vadedeki amacı, CT görüntüleme ile defekt bölgesinin belirlenmesinin ardından, hastanın kendi mezenşimal hücreleri ekildiği defekte uygun doku iskelesinin, toksik olmayan, kemiğin mekanik ve biyolojik özelliklerini sağlayan malzeme kullanılarak üretimi ile kişiye özgü doku mühendisliği alanına katkı sağlamaktır.
